Characterization of NAD-Dependent Formate Dehydrogenase from Trametes versicolor Using a Cell-Free Protein Expression System

CO2 emissions are the primary reason for global warming; hence, biological and chemical technologies converting into useful compounds being actively studied. Biological methods using enzymes can convert under mild conditions. Formate dehydrogenase (FDH) is a representative conversion enzyme. Its function was revealed after isolation from bacteria, yeast, plants. In this study, we evaluated potential of FDH isolated wood-rotting fungi. After isolating gene (TvFDH) Trametes versicolor, cloned full-length T. versicolor expressed it in cell-free expression system. The encoding TvFDH identified as 1,200 bp open reading frame (ORF) expected molecular weight protein approximately 42 kDa. Overexpression recombinant crude including confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Enzyme activities metabolite analyses efficiency reduction.
